I'm running the 64-bit version of CentOS 5.3. I followed the usual instructions for enabling Java in Firefox

cd /usr/lib/firefox*/plugins
ln -s /usr/java/jre1.6.0_14/plugin/i386/ns7-gcc29/libjavaplugin_oji.so

But it isn't working. Do I have to jump through any additional hoops to to java-enable firefox on a 64-bit version of CentOS?

to find out if a plugin is install go to address "about :plugins" in firefox .
i think you have a mistake here /usr/java/jre1.6.0_14/plugin/i386/ns7-gcc29/libjavaplugin_oji.so
it should be /usr/java/jre1.6.0_14/plugin/i386/ns7/libjavaplugin_oji.so

Actually, the 64-bit java plugin is <JAVA_HOME>/lib/amd64/libnpjp2.so assuming you installed SUN java. Plugin is in version 1.6.0u13 and greater.
